If I won the auction and paid for my item, can the seller cancel my purchase?

Apparently, they can! I have had that happen. The seller didn't get the price he/she wanted, so he/she cancels the sale and relists it. It shouldn't happen, but it does.

How do I cancel a bid after winning?

You can't, you bought it and it is a contractual outcome. You can ask the seller to cancel the transaction, but they do not have to and then you will get a non-paying bidder strike as the seller has to do that to get their money back because eBay has already charged them fees. The buyer can ask to cancel a sale if it is less than an hour after it.
