Windows recreates this file every time you plug in. So you actually delete the folder but Windows makes a new one.

If your flash drive or other device is write protected, there is nothing you can do (you need to be able to write to the drive in order to remove the folder). You could also try running CMD as administrator (search for CMD in start, right click, run as admin).
