Making a novel focusing on multiple characters amps up the difficulty since they have to be unique so they don't seem like carbon copies. So, they all have to be interesting since the story will focus on them and they cannot be bland side characters.

    If you mean that your novel takes place in the same world as the anime you're referencing, no. There could be copyright issues. If you mean that people in your world enjoy the anime in question, that's alright as long as it's subtle, and doesn't have any specifics, and is just a reference.
