Unless your 100 lb. cylinder has a "wet pipe," then forget it. (A wet pipe is one that goes down inside the tank so the end is always below the surface of the propane. It has a valve on the outside of the tank to which the other connections are made.)

How do I fill a new tank that has never been filled?

Connect it to the hose. Slowly open up the tank valve. Once the tank is 1 1/4 full, open up the bleeder valve. As soon as you see LP spraying out of the bleeder valve close it up, and close the main fill hose. Close up the tank. Release the pressure in the hose. Disconnect. Slap a propane only sticker on the tank. Call it a day.
