Endermen can teleport in, yes. This is always a possibility, so create a pumpkin farm. Spiders wont be able to get in unless you have a missing block somewhere. Be sure to have eaves so you can protect your roof.

It is a safe house because it protects you from hostile mobs. Zombies cannot get inside, unless it is hardcore, skeletons cannot get inside and creepers cannot get inside.

They can reach land and will follow you if you are near. However, if it is day, you don't have to worry since they burn up like zombies. At night just make sure everything is lit up with torches or lanterns, and then you'll be fine.
