You want to have activated the ? Switch. Once you have, go into Big Boo's Haunt as Mario, and go to the room on the second floor with the ? Block. Break the block, pick up the flower, and fly up to the ledge above it. Then enter the Luigi painting and go through the level as normal.

How do I unlock Luigi in Super Mario 64 DS?

To do this, you need over 15 stars. Then go downstairs till you find a big boos, follow it then kill all the boos till one drops a cage. You should be in big boo's haunt. you need to beat the first star. After, go upstairs then go to the book room find a wood stand a little above the ground. For this part to be easier, activate the ? switch then hit the block as Mario and press b over and over. You should find a high platform, then go through the door and go through Luigi's painting. if you don't activate the ? switch, keep trying to wall jump as Mario to the platform. It works because it is possible to beat the game and get every character without the ? blocks, it is just really hard.
