No, as the fish would probably be fine in the water. You should bring saltwater fish aboard in fish tanks, though, as rainwater is usually freshwater.

If the Bible is true, and Noah managed to build a boat of giant proportions in his era and lifetime, gather animals and prevent them from eating each other, two of almost every animal on earth, I don't see why he couldn't have stocked up on food as well and used rain barrels for water.

Do I need planning permission to build an ark?

If you are fined, say you will pay the fine 40 days and 40 nights after the completion of the ark. Seriously though, you should check in with your local municipality about any planning issues but if it's able to be moved and is not a permanent structure, you should be fine (not fined).
