You can explain that you feel better suited for the position you will now be assuming and thank your boss for the time and experience within the role you held with him/her.

If difficult coworkers are part of the reason I want to leave, should I mention this in my letter?

No. Thank your employer for the opportunities given to you and leave on a positive note. If you mention that difficult coworkers are part of the reason why you want to leave and a future employer contacts your old employer, your old employer may tell them you left because of workplace drama. That could go either way, and does not look attractive.
